腎臓、未分化マーカー、がんマーカー、糖タンパク質
1984年、ラットの糸球体に存在することが見いだされ、ヒトでは1986年に腎臓の糸球体から発見、1997年に同定された(Ref. 1, 2)。2003年にヒトEC細胞上のgp200という膜タンパク質と腎臓から発見されたポドカリキシンが同一のものであることが報告され(Ref. 3)、その後、ヒト及びマウスの血球芽細胞からも同定された。また、造血幹細胞マーカーであるCD34とアミノ酸配列に相同性をもつことで知られる。
一般的に膜タンパク質として存在し、タンパク質上に大きな糖鎖を有する。分子量はラットの腎臓では140kDa、サルおよびヒトの腎臓では170kDa程度である。ヒトEC細胞におけるタンパク質そのものの分子量は55kDaとされるが、通常150−200kDaを超える高分子として存在する。528個のアミノ酸に5箇所のN結合型糖鎖付加部位、3箇所のグリコサミノグリカン(枝分かれのない多糖)付加部位を有するほか、270個のアミノ酸によるムチンドメイン(O結合型糖鎖を多く付加する部位)を有する。未分化マーカーとして知られるTRA-1-60、TRA1-81などのプロテオグリカンのキャリアータンパク質でもある(Ref. 4)。
マウスおよびヒトの造血幹細胞から同定され、血液細胞の発育と分化に関与すると推測される。また、血管内皮からも同定され、白血球の血管外遊走に関与することも知られている。CD34関連ファミリーであり、内皮細胞接着レセプターであるL-セレクチンと特異的に結合する(Ref. 5)。相反するような2つの機能を持ち、一つは抗接着性を示し糸球体基底膜と隣接してポドサイトの濾過スリットの構造を作る。もう一つは、タンパク質相互作用を持ち、糸球体基底膜で血球と接着し濾過を行う(Ref. 6)。また、グルコーストランスポーターを細胞の正確な位置に方向づけし、その発現にも関与していると考えられる(Ref. 7)。
未分化な細胞で非常に多く発現し、分化とともに減少、特異的な抗体は分化抑制だけでなく細胞毒性を持つことが報告されている(Ref. 8)。また、未分化マーカーとしてだけでなく、様々ながんマーカー(前立腺、胸、肝臓、腎臓、甲状腺、胃、肺、大腸、脳、血液細胞)としても発現する。ヒトの精巣腫瘍のマーカーとして悪性の細胞に発現することが報告され、進行性のマーカーであるとされる(Ref. 9)。しかし、これらの機能としては、いまだ不明な点が多い
