インスリン、phosphatidylinositol 3-kinase (PI3-K)、高血糖
インスリンは糖、脂質、タンパク質の代謝作用のほか、細胞増殖、アポトーシスの抑制など、多岐にわたる生理作用を有する。インスリンの細胞内シグナルの経路のひとつにphosphatidylinositol 3-kinase (PI3-K)を介する経路があり、インスリンの代謝作用はこの経路の活性化によって発現する(Ref 1-3)。遺伝子はヒト11番染色体上に位置する。
SH2 domainを有する酵素で、1258個のアミノ酸からなる。
SHIP2は、脂肪組織や筋肉を初めとして生体に広範囲に存在し、PI3-Kの反応産物であるPI(3,4,5)P3を脱リン酸化し、インスリン作用を抑制する(Ref 4-6)。

SHIP2はインスリンの代謝作用を生理的に制御しているだけでなく、その発現の異常な亢進がインスリン抵抗性病態の一因に関与している可能性がある。自然発症 NIDDMモデル動物の一つであるdb/dbマウスの骨格筋および脂肪組織において、SHIP2の発現亢進が認められ、TZD系糖尿病治療薬の投与によるインスリン感受性の改善とSHIP2の発現の正常化が同時に起こることが報告されている(7)。我々はヒトSHIP2遺伝子の発現調節領域に存在するDNA多型と遺伝子発現との関係を解析し、5’UTRの変異(+334CC→CT,TT)はHbA1cが上昇していることを報告した(8,9)。SHIP2のノックアウト(KO)マウスに関する報告は複数存在し、インスリンに対する感受性が増し、激しい低血糖のために新生児時に死亡する(10)。他の報告では、SHIP2遺伝子の欠損は、生育には影響がなく、グルコースとインスリンのレベルは正常であったが、高脂肪食を摂取させた場合の体重増加に対して抵抗性がある(11)とのことである。我々が作成したSHIP2 transgenicマウスではむしろ、体重が低く耐糖能が維持されており、遺伝子発現レベルのみでなく、蛋白発現量やSHIP2の酵素活性そのものを測定する必要があると考える。
