長期記憶、長期増強(long-term potentiation; LTP)、認知症
cAMP-プロテインキナーゼA (protein kinase A, PKA)-CREB (cAMP response element binding protein) 系はほぼ全ての細胞に存在し、多様な機能を担っている。ATPから合成されたcAMPによって活性化されたPKAは、細胞中の酵素やCREBを始めとする転写因子をリン酸化する。CREB-1は、このcAMP-PKA-CREB系において遺伝子転写を促進する機能を有する。後の研究から、転写活性に必要なリン酸化部位を持たず、遺伝子転写において抑制的に働くCREB-1のアイソフォームが存在することが報告された (Ref 1,2)。
ヒトCREB-1遺伝子は第2染色体上に位置する (Ref 3)。CREB-1及びそのアイソフォームは、塩基性アミノ酸クラスターとロイシンジッパー領域からなるb-zip (basic leucine zipper) 構造を有している。これらの領域を介してホモ二量体あるいはヘテロ二量体を形成することにより、CRE (cAMP response element)に結合する。
CREB-1はほぼ全ての細胞に存在する遺伝子の転写因子であり、標的遺伝子のプロモーター領域上に存在するCREと結合することでPKAによるリン酸化依存的に遺伝子の転写を促進する
(Ref 4,5)。CREB-1によって遺伝子発現が促進される分子の中には、c-Fosやc-junなどの最初期遺伝子が含まれている (Ref
6)。これらの最初期遺伝子もまた転写因子として機能するため、CREB-1の関与によって多くの遺伝子発現が促進される。一連のカスケードによって発現した遺伝子及びタンパク質はシナプスにおける伝達効率の変化をもたらすため、細胞レベルでの記憶基盤と考えられている長期増強
(long-term potentiation, LTP) の誘発、維持に必要不可欠と考えられている(Ref 1)。
また、転写活性に必要なリン酸化部位を持たないCREB-1のアイソフォームが存在することも報告されており、遺伝子転写において抑制的に働くことが示されている
(Ref 1,2)。
CREB-1は短期記憶から長期記憶への移行に重要な役割を果たしていること (Ref
1,7)、またCREBが発現を誘導する遺伝子産物のうち、脳由来神経栄養因子や抗アポトーシス因子であるBCL-2が神経細胞の生存に寄与していることが報告されている(Ref
8)。また、老化あるいは細胞死や神経細胞の変性に端を発する種々の神経変性疾患に伴う記憶、認知機能改善のターゲットとしてcAMP-PKA-CREB系が注目されている
(Ref 9)。
近年ではCREB-1欠損マウスにおいてカロリー制限により活性化される長寿遺伝子SIRT-1(ヒストン脱アセチル化酵素)の発現が抑制されていることが報告されていることから(Ref
10)、CREB-1は老化に伴う多様な機能の低下に関与していると考えられる。
